Clomiphene citrate belongs to the triphenylethylene group and is classified as a selective estrogen receptor modulator with mixed agonist and antagonist properties. It acts as an antiestrogen by blocking the action of estrogen in most tissues. However, in bone tissue, it activates estrogen receptors, making it useful in preventing osteoporosis.
Mechanism of Action:
In men, clomiphene citrate acts as an antiestrogen by interfering with the negative feedback of estrogen on the hypothalamus. This stimulates increased production of gonadotropin-releasing hormone (GnRH), which in turn increases the production of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) by the pituitary gland. This cascade ultimately leads to increased testosterone synthesis and sperm production in the testes.
Clomiphene citrate acts on estrogen receptors in the pituitary gland and ovaries. By binding to these receptors, it interrupts estrogen’s negative feedback loop. Normally, estrogen reduces the pituitary gland’s production of FSH and LH. However, taking clomiphene citrate bypasses this inhibition. Clomiphene citrate does not directly affect blood estrogen levels. It acts as an estrogen antagonist, eliminating their inhibitory effect on the hypothalamus and pituitary gland, thus facilitating a more rapid recovery.
Usage Guidelines:
For post-cycle recovery, the dosage of clomiphene citrate can vary depending on the severity of the suppression. Typically, dosages range up to 150 mg in the first few days, gradually decreasing to 50-25 mg over two weeks. Adjustments can be made based on laboratory tests. For lighter cycles, such as those using oxandrolone alone, a dosage of 50 mg in the first week and 25 mg in the second week may be sufficient. However, it is essential to consider each case individually.
For testosterone-boosting therapy, a common daily dosage is 50-25 mg, with adjustments based on test results. Many men over 35 find a daily dosage of 25 mg satisfactory, experiencing increased testosterone levels, libido, and energy without side effects.
