Bremelanotide, also known as PT-141, is a compound developed for the treatment of female sexual dysfunction, hemorrhagic shock, and reperfusion injury. It activates melanocortin receptors in the central nervous system, modulating inflammation and limiting ischemia. Initially studied for intranasal use in female sexual dysfunction, development for this use was temporarily halted due to adverse effects on blood pressure. Subsequent studies explored subcutaneous injection, with minimal effects on blood pressure.
Bremelanotide is derived from the peptide hormone Melanotan 2, which has been studied for tanning without sun exposure. Melanotan 2 has been found to induce sexual arousal and spontaneous erections. Bremelanotide has been shown to be effective in treating sexual dysfunction in both men (erectile dysfunction) and women (sexual arousal disorder) by directly increasing sexual desire through its effects on the nervous system.
Action mechanism
Bremelanotide is an alpha-melanocyte-stimulating hormone (alpha-MSH) analogue that activates melanocortin receptors in the central nervous system. Its centrally mediated activity makes it potentially useful in the treatment of erectile dysfunction, decreased sexual motivation, and loss of libido.

Use
- The first erection occurs approximately 30 minutes after administration. Peak blood concentrations are reached within 60 minutes and gradually decline over the course of 4 hours.

Preparing the solution
To prepare a solution for injection, take a syringe with a diluent and inject it into a vial containing the lyophilized powder. Tilt the vial so that the needle touches the wall, allowing the diluent to flow slowly. Stir gently until the powder has dissolved and the solution is clear. Avoid mixing peptides in the same syringe.
Conservation
- The solution can be stored in the refrigerator at 2 to 8 degrees Celsius for approximately 21 days. If using bacteriostatic water, the shelf life may be longer.
